Transter of RP4::Mu cts and RP4::mimi-Mu from E. coli to Pseudomonas sp.

Abstract

Cbromosomal gene transferable hybrid plasmids, RP4::Mu cis and RP4::mini-Ma, were transferred by conjugation from E. coil to Pseudomonas strains. In order to use for recipient cuts of RP4:: Mu cts and RP4:: mini-Mu, pbum d-free Psadomonar strains were characterised for their antibiotic resistance, aromatic hydrocarbon utility and degradation patterns of chlorinated berbidde. Transfer frequencies of RP4:: mini-Ma exhibited about 10-2 to 10-4, while those of RP4:: Mu cts exhibited very low value of lo¢¥7 in recipients tested except Psadomonas aerwgnosa KU557. Existaace of hybrid plasmids In Pseudomonas transcoijugants were Identified by their antibiotic resistance and agarose gel dectrophoresls. In case of RP4:: Ma cts trancon ju`aats it was also confirmed by demoustrating that they were capable of releasing phnge and form rag plaques at 43¡ÆC. Plaque forming unit of the transcoojugants was about 105. It was shown by the stability test that RP4:: Mu cts and RP4:: ml lMu In Pseudomonas were relatively stable.
